For years, identical twins posed one of forensic science's toughest puzzles. Because they share nearly the same DNA, a standard genetic match could point to both siblings at once, leaving investigators without a clear answer.

That limitation is now shrinking. Scientists have learned to search for tiny mutations that can appear after an embryo splits into two. These rare differences may be present in one twin but not the other, creating a more precise genetic signature.

A new layer of DNA analysis

By using whole-genome sequencing, ultra-deep sequencing, and other advanced methods, researchers can scan billions of DNA letters to find those subtle variations. In some studies, the differences were strong enough to distinguish one twin from another with remarkable statistical confidence.

The approach has already moved beyond the lab. In several investigations, forensic teams used these methods to separate identical twins in cases where conventional DNA testing could not. One landmark U.S. case in 2025 became the first known conviction based on this kind of twin-specific genetic analysis.

The science is still demanding. It requires enough biological material, careful validation, and a mutation that is also present in the crime-scene sample. Even so, the field is advancing quickly, and courts are beginning to recognize its value.

Forensic genetics is entering a more detailed era, where even twins can leave distinct molecular traces. In the future, this could make DNA evidence more precise, more informative, and far more powerful in complex investigations.
