First identified in 2015, MOTS-C is unusual because it is encoded in mitochondrial DNA rather than in the nucleus. Researchers believe it may influence the body in ways that resemble the benefits of exercise, especially through the activation of AMPK, a key regulator of cellular energy.
When AMPK is activated, cells can improve glucose uptake, increase fat metabolism, and support mitochondrial performance. Early findings, including animal research and limited human data, suggest MOTS-C may help address insulin resistance and diet-related weight gain. Some studies have also reported improvements in gait and grip strength.
The peptide is now being studied in a 12-week clinical trial involving adults with obesity and prediabetes. At the same time, the U.S. Food and Drug Administration is reviewing MOTS-C among several peptides under consideration for compounded medicine pathways, with an advisory committee meeting planned for 2026.
While MOTS-C is not approved for general human use, its growing scientific profile reflects a broader shift toward therapies that target metabolism at the cellular level. If ongoing research confirms its potential, it could open a new chapter in precision approaches to obesity and longevity.
